DNA Sequencing Sample Preparation Guidelines
Updated April 2, 2008

All sample preps should only contain, DNA, primer (if added), and water. Please avoid using any buffers, which can interfere with the sequencing reaction, purification, and injection on the instrument ultimately diminishing data quality. Qiagen mini or maxi preps are recommended purification kits.

NEW SERVICE: The Difficult Template service is recommended for those wishing to sequence hairpins, GC-rich templates, or any sample that is being "difficult" and requires special attention. Difficult Template samples require additional information that should be supplied in the comments section of the online order form.

Primer Added Fast Track (you add primer) $8

Plasmid
50 ng/ul DNA
3.2 pmol primer already added
>12ul volume of DNA plus primer per reaction (we need at least 600ng of DNA per reaction)

PCR
5ng/ul DNA
3.2 pmol primer already added
>5ul per reaction (we need at least 25ng of DNA per reaction)

Primer NOT Added Fast Track (we add primer) $10

Facility Primers

Plasmid
50 ng/ul DNA
>12ul per reaction (we need at least 600ng of DNA per reaction)
*Primer is NOT already added and either supplied in a separate tube or stored at MBCF

PCR
5ng/ul DNA
>5ul per reaction (we need at least 25ng of DNA per reaction)
*Primer is NOT already added and either supplied in a separate tube or stored at MBCF
